No, I don’t think so. Make your site be in compliance with google adsense and you will get approval

No. But when you have a custom domain, your approval rate is higher than when you don’t.

No but If you don’t have a custom domain, you’ll get a hosted adsense account. You won’t be able to add other sites to the account or change the domain that got you the Adsense account. You will forfeit the account and all earnings when you get a custom domain.
